objective: objective: dermal papilla and hair epithelial stem cells regulate hair formation and growth cycle. damage to or losing of these cells can cause hair loss. although several studies have claimed to reconstitute hairs using rodent cells in animal model, a stable human hair follicle reconstitution protocol needs more studies. here in this study we evaluated hair induction through injecting adult cultured human dermal papilla cells and a mix of hair epithelial and dermal papilla cells in mouse model. materials and methods: materials and methods: discarded human scalp skins were used to obtain dermal papilla and hair epithelial cells. after separation, cells were cultured and assessed for their characteristics. fifteen c57bl/6 nude mice were randomly allocated to three groups to receive injections in dorsal skin. the first group they received cultured dermal papilla cells, the second group was received mixture of cultured epithelial and dermal papilla cells, and the third group was control group (pbs-). results: results: histopathologic examination from injection sites showed evidences of hair growth in all of the samples that had received cells in comparison with control group; however, mix group showed evidences of hair growth in examination by unequipped eye. pkh tracing evaluation confirmed the presence of transplanted cells in new hair. conclusion: conclusion: our data showed that injecting combination of adult human cultured dermal papilla and epithelial cells could induce hair growth in nude mice. this study emphasizes that the combination of human adult cultured dermal papilla and epithelial cells can induce new hair in nude mice.
